Zanidatamab with chemotherapy, with or without tislelizumab, for previously untreated locally advanced or metastatic HER2-positive gastroesophageal adenocarcinoma

Zanidatamab with chemotherapy, with or without tislelizumab, is in clinical development as a first-line treatment option for adults with unresectable, locally advanced, or metastatic, human epidermal growth factor receptor 2-positive (HER2+) gastroesophageal adenocarcinoma (GEA). This cancer starts when abnormal cells in the food pipe and stomach divide and grow in an uncontrolled way. Overproduction of a protein called HER2 can cause cancer cells to divide uncontrollably, forming a tumour. Advanced or metastatic GEA means that a cancer that began in the food pipe or stomach has spread to at least one other part of the body. Risk factors include smoking, obesity, acid reflux, diet, gender, older age, alcohol consumption, and long-term bacterial (Helicobacter pylori) infection. Current therapy involves a combination of targeted therapy with chemotherapy, but there still remains an unmet need.
