Selinexor with ruxolitinib for treating myelofibrosis

Selinexor with ruxolitinib is in development for the treatment of Janus kinase inhibitor (JAKi) treatment-naïve myelofibrosis in adults. Myelofibrosis is a rare type of blood cancer with poor survival rates. JAK is a type of protein that regulates cell growth, and becomes abnormally active in myelofibrosis, and can be treated with JAKis. JAKi treatment-naïve means the patient has not been treated with any other prior JAKi. Myelofibrosis mainly affects older people and is caused from multiple spontaneous changes in a person’s bone marrow DNA. Ruxolitinib is a commonly used therapy but many patients either become resistant to it or stop using it because of negative side effects.
