Secukinumab for treating polymyalgia rheumatica in recently relapsed patients

Secukinumab, given in combination with a glucocorticoid taper regimen, is in development for the treatment of patients with polymyalgia rheumatica (PMR) who have recently relapsed whilst
undergoing a glucocorticoid taper. PMR is characterised by inflammation of the muscles, most commonly presenting in the muscles around the shoulders, neck and hips, which causes pain and
stiffness, especially in the mornings. The incidence of PMR increases with age and is more likely to affect women than men. Although the exact underlying causes of PMR are unknown, it is
believed that a combination of genetic and environmental factors may be responsible. Currently PMR is treated through the use of glucocorticoids, with a gradual reduction in dose over time if
there are no recurrences of symptoms. However, around 43% of patients with PMR will experience at least one relapse within a year of treatment initiation, and there are common side effects to extended periods of glucocorticoid use.
