Ipatasertib with fulvestrant for advanced HER2-negative and ER-positive breast cancer

Ipatasertib with fulvestrant is in development for treating advanced human epidermal growth factor receptor 2 (HER2)-negative and oestrogen receptor (ER)-positive breast cancer after treatment with a CDK 4/6 inhibitor and aromatase inhibitor. Advanced cancer includes cancer that has grown outside the part where it started (locally advanced) and when it has spread to other parts of the body such as liver, bones, lung (metastatic). If the breast cancer is ER-positive, it means the cancer cells grow in response to high levels of the hormone oestrogen. HER2 is a protein that helps breast cancer cells grow quickly. Breast cancer that is HER2-negative means it has zero to normal levels of the HER2 protein. The main symptoms of breast cancer include: lump or swelling breast, bloody nipple discharge, breast or armpit pain. It is not always clear what causes breast cancer, but people are more likely to get it if they are: over 50 years old; have dense breast tissue; have a family history of breast or ovarian cancer; have certain other breast conditions; and if they have higher levels of the hormones oestrogen, progestogen or testosterone.
