Giredestrant with everolimus for HR+/HER2- advanced breast cancer after CDK4/6 inhibitors and endocrine therapy

Giredestrant with everolimus is in clinical development for the treatment of oestrogen receptor positive (ER+), human epidermal growth factor receptor 2-negative (HER2-), locally advanced or metastatic breast cancer after cyclin-dependent kinases (CDK) 4/6 inhibitors and endocrine (hormone) therapy. Breast cancer occurs when abnormal cells begin to grow uncontrollably and form a tumour. Locally advanced breast cancer has spread from the breast to areas close to the breast or to the chest wall. Metastatic cancer has spread to other parts of the body. ER+ cancer has receptor cells for the hormone oestrogen, meaning that oestrogen stimulates the cancer cells to grow and divide. This type of cancer often responds to treatment with endocrine therapy. However, these treatments have significant impacts on quality of life which can greatly affect treatment adherence and ultimately, patients can become resistant to them. Prolonged treatment and risk of relapse can represent significant challenges for patients with breast cancer and therefore, a need remains for more effective and acceptable treatment options.
