Encaleret for autosomal dominant hypocalcaemia type 1

Encaleret is currently in development for the treatment of autosomal dominant hypocalcaemia type 1 (ADH1). ADH1 is an inherited condition caused by a change in one copy of a gene called the calcium-sensing receptor gene (CASR). The CASR gene controls a sensor in the body called the calcium-sensing receptor (CaSR) that helps keep calcium levels in the blood normal. Certain changes in this gene make the sensor too sensitive, so the body thinks there’s more calcium than there really is. This causes low blood calcium, which can lead to tingling, muscle cramps or spasms, twitching, stiffness, and seizures. Treatment usually includes calcium and vitamin D, but this can sometimes cause too much calcium in the urine, which may harm the kidneys over time.
