Durvalumab with oleclumab or monalizumab for treating locally advanced, unresectable non-small-cell lung cancer

Durvalumab with oleclumab or monalizumab is in development for the treatment of locally advanced, unresectable (non-operable) non-small cell lung cancer (NSCLC) which has not progressed following platinum-based chemoradiation. Symptoms of NSCLC can vary, with common symptoms including new or persistent coughs, getting out of breath doing easily, coughing up blood, and aches or pains in the chest. The largest risk factor for lung cancer is smoking, however environmental hazards such as silica or engine fumes also increase risk. Durvalumab is currently a treatment for NSCLC when platinum-based therapies have been insufficient and has been shown to increase survival rates, however many patients fail to respond to treatment or develop a resistance. It has been proposed that combining durvalumab with oleclumab or monalizumab may provide a novel approach to further improve patient outcomes by targeting alternate signalling pathways that have shown potential antitumor activity.
